Change opacity of section on image

Image is 800px x 150px, i want to change the opacity in the top left of the image for a rectangular section of 300px x 50px.

So i thought i would highlight the section(300px x 50px) with the rectanglar marquee tool, which gives a dotted rectangle, now i thought by changing the opacity slider over on the right side in the layers palet then this would work, but instead the opcaity for the whole image is still changed, how do i have only the section i want to change the opcaity for ?

Not sure what version of PS you are running. This is based on CS4. However, it shouldn't be to difficult to follow in earlier versions as I think most of the shortcuts I mention here are the same in previous versions.

Make sure you have an empty layer below all others. If you are editing an existing image, the background layer may be locked. There are several ways to unlock it. One is to right click on the background layer in the layers pallet and select Layer from background. After it is unlocked, create a blank layer under it by holding Ctrl and clicking the new layer button at the bottom of the layer pallet. Make the original layer active and make your selection for the area you want transparent. Hit Ctrl+X to cut that area from the original. Hit Ctrl+Shift+D to reselect. Hit Ctrl+V to paste the part you cut onto a new layer in the exact position it was in. Adjust the transparency level of that layer.
