Hello,
I'm sure this is quite difficult to achieve realistically but what is the best way to go about removing the clear plastic stand from a picture of a ring and then repainting a metal band below? Also any advice on touching the shots up, I already made some changes to the levels.
Please see the pictures below for some examples.
Any help would be greatly appreciated!

Bad news, unless you can get a matching photo of the inside of the ring with no plastic stand in the way it will take some time and effort to fix.
Use the elliptical marquee tool to create masked areas to easily remove the plastic holder. Then using the same elliptical marquee tool create selections to create the area you need to fill in. Think of the different parts of the ring as separate areas. The side, the inside, the back edge, etc. You will need a combination of the elliptical marquee tool with Add to selection and Subtract from selection to create the correct mask. Make sure you save those selections so that you can easily go back to them. Then it is a matter of using your paint tools, clone stamp tools, airbrush tool, etc to paint in the missing part of the ring. With some careful work you can replace it so that no one will spot it.
